Current cancer immunotherapies often show limited efficacy due to suppressive tumor immune microenvironments (TIMEs). To overcome this, localized delivery of tumor antigens and immune stimulants is essential. However, repeated administrations can be inconvenient and lack precise immune control, increasing the risk of cytokine storms. Here, we present a patient-adaptable nano-gel-nano platform enabling sustained generation of tumor-associated antigens and release of adjuvants. This is achieved through the specific delivery of drugs to cancer cells and antigens/adjuvants to immune cells over the long term, maintaining proper concentrations within the tumor site with a single injection. This system maintains effective concentrations within the tumor via a single injection, supports tumor-specific immunity, enhances immune checkpoint blockade efficacy, and induces systemic memory T cell responses, while minimizing systemic toxicity.
