Nitroreductase and Reduction Responsive Ion Pair Self-Assembled Nanocarriers Based on Poly(diallyldimethylammonium chloride) and 5,5′-Dithiobis(2-nitrobenzoic acid)

An ion pair self-assembled micellar (IPSAM) nanocarrier was developed using poly(diallyldimethylammonium chloride)(PDADMAC) and 5,5′-dithiobis(2-nitrobenzoic acid)(DTNB). These two compounds formed electrostatic ion pairs via interactions between amino and carboxyl groups, resulting in amphiphilic complexes that spontaneously self-assembled in water. IPSAMs at molar ratios of 5/5, 6/4, and 7/3 (amine/carboxyl) formed stable white suspensions. Ion-pair formation was supported by characteristic FT-IR and NMR shifts. TEM confirmed micellar morphology, and hydrodynamic diameter, zeta potential, and CMC were measured. Upon DTT treatment, the UCST decreased with increasing dithiothreitol(DTT) concentration, showing redox responsiveness. Nitroreductases(NTR) treatment did not alter UCST but increased transmittance. Both stimuli enhanced the release of Nile red, indicating the system's reduction and nitroreductase responsiveness.
