Synthesis of a Zwitterionic Chitosan and its Antifouling Coatings for Medical Devices

Peripherally inserted central catheters are widely used for prolonged intravascular therapies, including blood transfusion and drug delivery. However, extended use increases the risk of severe infections and thrombosis. Zwitterionic polymer coatings offer a promising strategy by forming a highly hydrated antifouling layer that provides antibacterial properties while reducing thrombosis risk. The hydration layer inhibits coagulation factor adhesion, and electrostatic repulsion further minimizes thrombogenesis. In this study, we synthesized a zwitterionic polymer by conjugating isonicotinic acid (INA) with 1,3-propanesultone to produce 3-(4-carboxypyridin-1-ium-1-yl)propane-1-sulfonate (SB-INA), which was then conjugated to chitosan. The resulting zwitterionic chitosan polymer was coated onto a medical titanium substrate, and its antifouling properties were evaluated. This presentation provides our strategy for designing a zwitterionic chitosan derivative and its biomedical applications.
