Structural and Rheological Characterization of Triblock Copolymer-Based Hydrogels During Hydrolysis

Poly(D,L-lactide) (PLA) is an FDA-approved polymer for drug delivery systems, and with a combination of hydrophilic polyethylene oxide (PEO), PLA-b-PEO-b-PLA triblock copolymers offer excellent biodegradability and biocompatibility. In this study, we synthesized a PLA-b-PEO-b-PLA triblock copolymer and evaluated the structural and rheological properties of hydrogel made from simple aqueous self-assembly of the polymer. The copolymers were synthesized via ring-opening polymerization with Tin(Ⅱ) 2-ethylhexanoate(Sn(Oct)2) as catalyst.
The degradation behavior of the hydrogels was monitored over time under physiological temperature 37℃.
The hydrogels exhibited time-dependent degradation with observable changes in structural and viscoelastic properties (e.g., PLA domain size, dynamic moduli). These results suggest the potential of PLA-b-PEO-b-PLA based hydrogels as stimuli responsive plat forms for controlled drug delivery applications.
