Magnetically Guided Amine-Functionalized Hydrogel Beads for Targeted Delivery of Doxorubicin in Transarterial Cancer Chemoembolization

Gelatin methacryloyl (GelMA), synthesized via methacrylation of gelatin with methacrylic anhydride (MA), offers outstanding biocompatibility and biodegradability for biomedical applications. By employing water‑in‑oil emulsion polymerization, GelMA was copolymerized with 3‑sulfopropyl methacrylate (SPMA) to generate drug‑eluting GelMA‑SPMA hydrogel beads. In transarterial chemoembolization (TACE) for hepatocellular carcinoma, electrostatic attraction between doxorubicin and the sulfonate groups of SPMA resulted in markedly enhanced drug loading in the beads. To enable magnetic targeting and precise intravascular positioning, superparamagnetic iron oxide nanoparticles (SPIONs) were incorporated into the beads. Amine‑functionalized SPIONs demonstrated superior magnetic responsiveness compared to pristine SPIONs.
