Bioinspired Gradient Hydrogel Scaffold for Accelerated Skin Wound Healing

Hydrogel scaffolds are widely utilized in tissue engineering however replicating the complex microenvironment of native tissue remains challenging. In this study a gradient hydrogel incorporating L929 mouse fibroblast spheroids was developed to better mimic the mechanical properties of skin and enhance wound healing. The gradient was achieved by modulating the concentration of methacrylated hyaluronic acid (HA-MA) via a dual-syringe pump system. The resulting hydrogel facilitated organized cell behavior and significantly promoted full-thickness skin regeneration. In vivo studies demonstrated that treated wounds exhibited a 1.53-fold increase in healing rate compared to untreated controls. These findings highlight the potential of the gradient hydrogel as a biomimetic scaffold for advanced wound healing and tissue engineering applications.
