Nonspecific protein adsorption decreases biosensor accuracy. Hydrophilic or zwitterionic surface modifications can reduce fouling, but their instability and complex synthesis limit use. Here, we present a simpler yet effective alternative: a fluorinated polymer-based hydrophobic coating. A copolymer was synthesized from 2,2,2-trifluoroethyl methacrylate (TFEMA) for low surface energy and hydrophobicity, and n-butyl acrylate (BA) for flexibility and strong adhesion. The copolymer can be applied to diverse substrates via a simple one-step spin-coating process. The coating showed excellent antifouling against proteins and cells, with thickness precisely controlled by polymer concentration to optimize performance. This coating can be stably and reproducibly applied to various substrates regardless of material type. Our work offers a robust, facile, and versatile surface modification strategy with strong potential for antifouling applications in biosensing platforms.
