Nanosheet-Engineered Tumor Spheroids for Enhanced Simulation of the Tumor Microenvironment

Conventional cancer models often fail to replicate the complex tumor microenvironment (TME), limiting nanotherapeutic evaluation. This study introduces a nanosheet-based tumor spheroid platform that better recapitulates in vivo tumor characteristics. Nanosheets enable uniform 3D spheroid formation with enhanced intercellular adhesion and structure. These spheroids demonstrate concentration-dependent increases in hypoxia and in reinforced cell–extracellular matrix (ECM) interactions. Notably, fibronectin expression is upregulated, promoting ECM remodeling and TME architecture. This enhanced microenvironmental simulation enables more accurate evaluation of nanoparticle-based therapeutics. Recapitulating key pathophysiological features of solid tumors, this nanosheet-integrated spheroid system provides a robust platform for preclinical screening of nanomedicines. This approach demonstrates significant promise to advance precision cancer therapeutics and translational outcomes.
