Adiponectin-Derived Peptide-Conjugated Heparin/Chitosan Hydrogel for Keloid Scar Treatment

Keloid scars are characterized by excessive fibroblast activity during the wound healing process, leading to overproduction of collagen. While various strategies have been employed to treat keloid scars, they are often associated with pain and undesirable side effects. In this study, we developed a biocompatible hydrogel physically crosslinked with heparin and chitosan, incorporating the adiponectin-derived peptide ADP355, for keloid therapy. ADP355, known for potential therapeutic effects in liver fibrosis and systemic sclerosis, activates AMPK and subsequently inhibits TGF-β1 signaling, showing promising antifibrotic effects in keloids. To enhance its bioavailability and therapeutic efficacy, ADP355 was conjugated with heparin and subsequently embedded in a heparin/chitosan hydrogel matrix. This hydrogel-based approach offers a promising, minimally invasive alternative for the effective treatment of keloid scars, with reduced pain and side effects.
