Engineering a Photosensitizer for Precision Modulation of Biopolymer Condensates

Recent studies show that specific biomolecular condensates are crucial in cancer initiation and progression and are linked to drug resistance, making them increasingly important therapeutic targets. In this study, we designed and synthesized an iridium-based photosensitizer capable of targeting biomolecular condensates. Furthermore, this neutral iridium-based molecule as a biocompatible photosensitizer with low dark toxicity offers the advantage of minimizing side effects. The photosensitizer enables the spatiotemporal generation of reactive oxygen species (ROS) upon light irradiation. By controlling light intensity, the oxidative stress induced by these ROS can be regulated, allowing for diverse condensate transformations in a precise manner. This system presents a novel approach to modulating condensate dynamics. In conclusion, this study investigates cancer cell changes induced by condensate modifications, potentially broadening its application as a novel therapeutic strategy.
