Development of a PHB/PEO-Based Electrospun Fiber Sheet to Prevent Lung Air Leakage.

Pulmonary air leaks after lung resection can cause infection and cardiopulmonary complications. To address this, an ultrafine fiber sheet composed of poly(3-hydroxybutyrate) (PHB) and polyethylene oxide (PEO) was developed. Mimicking the extracellular matrix, the sheet offers high surface area and porosity, enhancing biocompatibility. Although PHB is biodegradable and biocompatible, its low crystallinity can lead to fiber fusion during electrospinning. Incorporating PEO improved fiber uniformity and crystallinity, as confirmed by FT-IR and DSC. Water contact angle analysis showed increased hydrophilicity with higher PEO content. Tissue adhesion, measured by rheometer also increased with PEO concentration. Burst pressure tests demonstrated effective air sealing. The PHB/PEO (7:3) sheet showed rapid degradation and excellent cytocompatibility in vitro. Histological analysis revealed superior skin regeneration compared to gauze and NEOVEIL™. These findings suggest that PHB/PEO sheets are promising candidates for sealing pulmonary air leaks.
