cECM-incorporated Bioactive Hydrogels for Burn Wound Healing

Burn wound healing continues to present a clinical challenge owing to extended inflammation and the lack of a conducive microenvironment. To address these limitations, various biomaterials have been developed. Among these, cell-derived extracellular matrix (cECM)-based hydrogels have demonstrated potential in modulating cellular behavior, directing macrophage polarization, and offering a bioactive microenvironment. Here, we develop cECM-incorporated bioactive hydrogels (Gel-cECM). Gel-cECM demonstrate adequate mechanical strength and preserve ECM components, providing structural support and supporting long-term cell-matrix interactions. In addition, Gel-cECM enhance host cell infiltration, M2 macrophage polarization, angiogenesis, and collagen deposition in vivo. Notably, they accelerate wound healing and the regeneration of skin appendages, including hair follicles, muscle fibers, and nerves. In sum, Gel-cECM offer significant potential for burn wound healing and tissue regeneration.
