Antigen Presenting Cell Mimetic Lipid Nanoparticles Loaded with mTOR siRNA for Regulatory T cell Induction

Regulatory T cells (Tregs) are immunomodulators that reduce inflammation and promote tissue regeneration. Recently, inducing Tregs has emerged as a promising strategy for tissue regeneration. This process requires initial activation of naive T cells by co-stimulation of CD3 and CD28. Also, the downregulation of the mammalian target of rapamycin (mTOR) pathway has been implicated in promoting Tregs induction. In this study, we developed lipid nanoparticles (LNPs) conjugated with CD3e and CD28 antibodies and loaded with mTOR-siRNA (Ab-LNPs). We assessed the activation efficiency of Ab-LNPs by measuring the enhanced expression of CD25, a key marker of T cell activation. Moreover, the Ab-LNPs exhibited higher siRNA delivery efficiency compared to control LNPs. These findings suggest that the Ab-LNPs effectively promote the activation and induction of Tregs from naïve T cells, offering a potential therapeutic strategy for tissue regeneration.
