Nucleic acid-based micelle incorporated with USE1-silencing siRNA for lung cancer therapy

UBA6-specific E2 conjugation enzyme 1 (USE1) is frequently overexpressed in lung cancer and has been implicated in tumor progression. To develop a targeted RNA interference (RNAi)-based therapeutic, we introduce U4T, a lipid-modified DNA carrier composed of four consecutive dodec-1-ynyluracil residues. U4T self-assembles into micelles and forms a soft spherical nucleic acid aggregate (siSNA) through simple hybridization with siRNA targeting USE1. The resulting siSNA demonstrates efficient cellular uptake without the need for cationic transfection agents and exerts potent anti-tumor effects. Specifically, siSNA significantly inhibits cell proliferation, migration, and invasion, induces G1 phase cell cycle arrest, and promotes apoptosis in lung cancer cells. In vivo studies further confirm its ability to suppress tumor growth. These results highlight siSNA as a promising and clinically applicable RNAi-based nanotherapeutic for lung cancer treatment.
