Type I antifreeze protein-DNA hybrid nanostructures for ice recrystallization inhibition

Antifreeze proteins (AFPs) protect organisms that inhabit a sub-zero environment from freezing. The type I AFP, isolated from the fish species, contains high alanine content and adopts an α-helical structure with cap at the carboxy termini. Here, we report the peptide-DNA hybrid structure(PDH) in which DNA tethered to the end of type I AFP. We designed a peptide nucleic acid(PNA) of 6 bases for connecting DNA and peptide. The PDH structures were compared to AFP activity by their inhibition of ice recrystallization(RI). We calculated the α-helical formation rate of the PDH structures and the molecular modeling of 1WFA conformational changes depending on the PNA binding site.
