Surface Charge-Dependent Uptake of Pluronic Nanoparticles by HeLa Cells

Surface charge is a key factor influencing nanoparticle (NP) interactions in biological systems. To investigate its role in cellular uptake, we synthesized Pluronic block copolymer F127-based NPs functionalized with phosphate (PP) to impart negative charge and trimethylammonium bromide (AB) to confer positive charge. By varying the ratios of PP-F127 and AB-F127, we prepared self-assembled NPs with tunable surface charge densities and a semi-interpenetrating core structure. These NPs were characterized and evaluated for cellular uptake in HeLa cells. The results demonstrated that positively charged AB-NPs exhibited significantly enhanced cellular internalization compared to negatively charged PP-NPs, underscoring the critical role of surface charge in NP-cell interactions. These findings provide valuable insights into the design of NP-based drug delivery systems by highlighting the influence of surface charge on cellular uptake
