Surface-Modified Biochips by Polymeric Thin Film for Multiplexed and Flexible Biosensing

In this study, we aimed to construct a versatile biosensing platform using surface modification strategies. Previously the optical detection in biosensor modification exhibited limitations due to its sensitivity to chip thickness, transparency and geometry, restricting the platform's structural flexibility. To overcome these constraints, we introduced both organic and inorganic materials to functionalize surfaces capable of selective biomolecules such as nucleic acid, protein or carbohydrate capture. For the enhanced RNA detection, we modified the chip surface as a positively charged polymer coating, and we developed polymer capping layers on a copper electrode biochip to enhanced stability and sensitivity of a non-enzymatic glucose sensor. These approaches demonstrate that nanoscale blended polymer thin films can support both optical and electrochemical biosensing in multiplexed biomarker detection, POCT (Point-of-Care Testing), and wearable diagnostics.
