Three-dimensional hyaluronic acid hydrogel with low-intensity ultrasound stimulation accelerates cellular reprogramming efficiency into induced pluripotent stem cells

Induced pluripotent stem cells (iPS) present significant potential for tissue regeneration due to pluripotency, but traditional methods have low efficiency. Previous study showed that hyaluronic acid (HA) hydrogel enhances iPS generation. In this subsequent study, we explored whether low-intensity ultrasound stimulation (LIUS) improves iPS generation in HA hydrogel. Notably, we observed a time-dependent enhancement in iPS generation following LIUS, validated through OCT4-GFP intensity, colony formation, and pluripotency markers. LIUS also increased CD44 expression and membrane fluidity while collapsing cytoskeletal structures. LIUS also promoted epigenetic modifications, as evidenced by elevated levels of Acetyl H3, H3K4me2, and H3K4me3 markers. Importantly, 3D HA-LIUS system also achieved successful iPS generation in vivo. To summarize, LIUS as a 3D external stimulus would be a useful platform technique for achieving massive production of iPS by fostering cellular reprogramming.
