Epigallocatechin Gallate–incorporated Carboxylated Poly-L-lysine Solution for Cryopreservation of Stem Cell Spheroids

Despite the therapeutic potential of stem cell spheroids, their distinct structural characteristics require optimized cryopreservation strategies, and conventional protocols often fail to maintain high post-thaw viability and integrity. To address this, we developed a cryopreservation solution incorporating epigallocatechin gallate (EGCG), a natural antioxidant extracted from green tea leaves, into carboxylated poly-L-lysine (COOH-PLL) to create an EGCG-loaded COOH-PLL (E-CPLL) system. The physicochemical properties of E-CPLL were analyzed according to the variation in functional groups. Human mesenchymal stem cell (MSC) spheroids preserved with E-CPLL exhibited significantly improved post-thaw viability, antioxidant activity, and structural stability compared to those treated with conventional cryoprotectants such as dimethyl sulfoxide (DMSO). These findings suggest that E-CPLL is a promising candidate for spheroid cryopreservation, minimizing cell death through synergistic chemical and biological protection.
