Thermogelling Octanoyl Glycol Chitosan/Dialdehyde-terminated PEG Hydrogels with Enhanced Mechanical Properties Via Dynamic Schiff-Base Crosslinking

Abstract

Designing injectable hydrogels that combine shear-thinning, rapid recovery, and tunable mechanical properties is vital for biomedical use, yet maintaining both fast gelation and long-term stability in dynamic systems remains a key challenge. In this work, we report a hybrid hydrogel formed by integrating thermogelling octanoyl glycol chitosan (OGC) with dialdehyde-terminated poly(ethylene glycol) (DP), yielding dual functionalities of thermal responsiveness and reversible covalent crosslinking. The resulting hydrogel showed excellent injectability, rapid gelation, self-healing capacity, and mechanically tunable behavior, primarily governed by dynamic Schiff-base bonding and physical interactions. Fine-tuning of the DP composition enabled precise control over the hydrogel properties. These features support its promising role in drug delivery systems and tissue engineering scaffolds.
