Unraveling the mechanism of nitric oxide release for prolonged sustained release in PLGA-mediated delivery

Abstract

Diabetic foot ulcers (DFUs), a major diabetes complication, involved poor angiogenesis and tissue necrosis, often leading to amputation. Current self-care based therapies are burdensome and poorly support regeneration. Nitric oxide (NO) promotes wound healing through immune modulation, angiogenesis, and antimicrobial activity. However, Its clinical use is restricted by limited stability, demanding a modulate release approach.
In this study, we developed PLGA shell-based NO delivery system designed to sustain NO release, therebt facilitating early-stage healing in DFUs. S-nitrosoglutathione (GSNO) was covalenty linked to PLGA and embedded in a hydrophilic core to mitigate initial burst release. Modulating the molecular weight of PLGA established a tunable diffusion barrier, enabling consistent and localized release of nitric oxide at therapeutically relevant levels. The system shows therapeutic promise for early diabetic wound treatment via targeted and prolonged nitric oxide release.
