Surface chemistry-dependent lipid adsorption variations across diverse silica nanoparticle coating platform

Abstract

Lipid coating enhances nanoparticle stability, skin permeation, and cellular targeting through surface modulation. Surface properties govern lipid–nanoparticle interactions, yet adsorption mechanisms remain poorly understood. We demonstrate lipid-coated hybrid nanoparticles via co-extrusion of lipids with functionalized silica nanoparticle. DLS reveals systematic changes in size and surface charge between lipid-coated hybrids, uncoated silica nanoparticles, and bare liposomes. Lipid film formation occurs across all surface chemistries, whereas TGA and DSC demonstrate surface chemistry-dependent lipid adsorption variations, attributed to curvature effects and hydration shell dynamics. NBD-labeled silica nanoparticles enable quantitative assessment of full-coating ratios. This framework guides epigallocatechin gallate (EGCG) encapsulation for targeted antioxidant delivery. These findings establish designs for surface-tailored lipid-coated nanoparticles in biomedical systems.
