Abstract
Cascade enzymatic reactions are key to complex biochemical processes but remain challenging to control in artificial systems. We report chiral plasmonic nanoparticles with glucose oxidase (GOD) and peroxidase (POD) activities for optically tunable catalytic cancer therapy. Using sequential circularly polarized light (RC then LC), we selectively activate GOD and POD, achieving 1.3× overall enhancement compared to uncontrolled cascades by optimizing the reaction environment. D-Au nanoparticles show 2× higher affinity for D-glucose due to chirality matching. In cell and mouse models, sequential irradiation yields the highest radical generation and best therapeutic efficacy. This platform offers strong potential for precision catalytic therapy.
