Abstract
Natural killer (NK) cells are key effectors in tumor immune surveillance, primarily eliminating cancer cells through antibody recognition. However, their therapeutic potential is hindered by heterogeneous tumor antigen expression. To address this, we developed a strategy to display Fc fragments directly on tumor cell surfaces using a synthetic fusion protein, Univody, composed of the human IgG1 Fc region fused to a transmembrane domain. The construct was delivered as plasmid DNA (pUnivody) via a phenylboronic acid (PBA)-functionalized lipopolyplex (LPP) system, facilitating targeted delivery while minimizing cytotoxicity. This approach enabled antigen-independent Fc display on tumor cells, leading to NK cell activation in vitro. In vivo, pUnivody@LPP-PBA significantly suppressed tumor growth in triple-negative breast cancer and melanoma models, demonstrating a versatile immunotherapy platform that circumvents tumor antigen dependence.
