Multi-shell structure upconversion nanoparticles-based multifunctional nanocarrier for combination cancer therapy

Abstract

Photodynamic therapy (PDT) is a minimally invasive approach for cancer treatment that relies on the generation of reactive oxygen species (ROS) through light-induced activation of photosensitizers in the presence of oxygen. However, its clinical efficacy is often hindered by limited tissue penetration of light, insufficient endogenous oxygen, and the modest therapeutic effect of monotherapy. To overcome these limitations, combination cancer therapies integrating multiple therapeutic mechanisms have gained significant attention. In this study, we developed multifunctional nanocarriers based on multi-shell structured upconversion nanoparticles (MUCs) for synergistic cancer therapy with PDT, chemodynamic therapy (CDT), and starvation therapy. The synergistic antitumor efficacy of the MUC nanocarriers was systematically evaluated in HeLa and A549 cancer cell lines, demonstrating enhanced therapeutic outcomes compared to individual treatment modalities.
