Lipid–PLGA Hybrid Nanoparticles for Enhanced Drug Delivery in Solid Tumors

Abstract

Drug delivery to solid tumors is challenged by hypoxia, acidic pH, and high interstitial pressure, which hinder drug penetration and therapeutic efficacy. This study investigates lipid–PLGA hybrid nanoparticles as a strategy to overcome these barriers. PLGA offers biocompatibility and controlled drug release, while the lipid layer enhances membrane interaction and cellular uptake, improving drug stability and intracellular retention.
Synthesized lipid–PLGA nanoparticles will be characterized for their physicochemical properties. To evaluate functional performance, a fluorescent model drug will be encapsulated to quantify drug loading efficiency and cellular uptake, comparing coated and uncoated PLGA nanoparticles. In addition, Gemcitabine, a chemotherapeutic agent, will be employed to assess therapeutic efficacy under tumor-mimicking conditions. This platform holds potential for future integration with diverse functional modules to address key delivery challenges in solid tumors.
