Abstract
Artificial cells are increasingly recognized as powerful tools for understanding biological complexity and building life-like systems. Among various platforms, proteinosomes, protein–polymer conjugate-based microcapsules, have attracted attention due to their self-assembly-driven formation, selective permeability and potential for enzymatic activity. However, achieving precise stimuli-responsive control remains a key challenge in efforts to mimic natural cell systems. Here, we introduce a new class of DNA-tethered proteinosomes that utilize DNA as dynamic linkers between building blocks. These microcapsules can be programmed to undergo structural changes in response to specific molecular cues. This strategy enables fine-tuned control over capsule integrity and presents a modular approach toward stimuli-responsive artificial cell systems. Our work highlights the potential of integrating DNA nanotechnology into soft matter-based synthetic cell platforms for programmable behavior.
