Designing Oxygen-Supplying Containers to Create Hyperoxic Hydrogels for Facilitating Wound Healing

Abstract
Chronic wounds remain challenging to manage due to their prolonged inflammation and complications. To address this, bioactive hydrogels have emerged as promising platforms for immune modulation by delivering bioactive molecules to the wound. Among them, oxygen (O2) plays a pivotal role in wound healing through immune modulation and tissue maturation. However, current O2-releasing hydrogels face critical limitations, including the cytotoxicity of O2-generating agents and their byproducts. Herein, we develop an O2-supplying container (Oxygener) using dopamine-mediated catalase immobilization to produce O2-enriched medium. By immersing hydrogels in this medium, we fabricate hyperoxic hydrogels capable of delivering O2 without any supplementary agents. Our hydrogels support tissue remodeling and promote the transition from pro- to anti-inflammatory phases in vivo. The Oxygener demonstrates great potential as a platform for advanced O2-delivering systems to facilitate wound healing.
