Abstract
We developed a tumor-targeted nanoplatform that induces ferroptosis and suppresses immune evasion. Iron oxide nanoparticles were coated with PEI to load PD-L1 siRNA and modified with hyaluronic acid for CD44-mediated targeting. This system selectively delivers therapeutics to cancer cells with minimal off-target effects. In vitro, treatment reduced cancer cell viability to 30% while sparing normal cells. Ferroptosis was confirmed by elevated ROS and lipid peroxidation. PD-L1 siRNA effectively silenced PD-L1 expression. In vivo, the nanoparticles inhibited tumor growth and accumulated in tumors, with low distribution to other organs. Immunohistochemistry showed decreased PD-L1 and increased CD8+ T cell infiltration. This nanoplatform integrates ferroptosis and immune checkpoint blockade for effective cancer therapy.
