Morphological and Compositional Engineering of Donor-Acceptor Nanoparticles for Enhanced Photodynamic Performance in the Hypoxic Tumor Microenvironment

Abstract

Conjugated polymer-based nanohybrids are promising phototherapeutic agents due to their tunable optoelectronic properties. In this study, nanoparticles were prepared using a modified phase separation method with D8PE-Ac and D8PE-PEG phospholipids, incorporating PCPDTBT as an electron-donating core material. Two structures were designed to examine the effect of acceptor location: (1) both PCPDTBT and Y6 distributed in the core, and (2) PCPDTBT in the core with Rose Bengal (RB) localized on the surface. Under 808-nm laser irradiation, reactive oxygen species (ROS) generation was analyzed to evaluate how acceptor positioning influences photodynamic efficiency. The results demonstrate that internal versus surface localization of acceptors critically affects ROS production. This study underscores the importance of molecular distribution within nanohybrids and offers insights for designing more effective nanostructures for cancer phototherapy.
