Scutellarein Attenuates Inflammatory Cytokine-Induced Atrophy in hiPSC-Derived Skeletal Muscle Cells by Preserving Myotube Structure and Myogenic Identity

Abstract

Cancer cachexia is a severe metabolic syndrome involving systemic inflammation and irreversible muscle wasting. We developed a human in vitro model using skeletal muscle cells derived from human induced pluripotent stem cells (hiPSCs) and tested scutellarein, a natural JAK2/STAT3 inhibitor, for therapeutic potential. hiPSCs were differentiated into mature myotubes over 46 days, providing a human-specific platform for disease modeling. Inflammatory atrophy was induced by TNF-α and IFN-γ, with scutellarein pre-treatment. Treated groups showed preserved myotube diameter, increased maturated expression, and improved sarcomeric integrity in a dose-dependent manner. These findings suggest scutellarein protects against inflammatory muscle atrophy and supports its potential as a cachexia therapy. The use of hiPSC-derived muscle cells offers a translationally relevant model for drug evaluation in human muscle disorders.
