Abstract
Upon exposure to biological environments, nanoparticles form a protein corona, causing opsonization and rapid clearance by the mononuclear phagocyte system, reducing therapeutic efficacy. Although polyethylene glycol (PEG) reduces protein adsorption, it cannot fully prevent immune recognition and may induce anti-PEG antibodies. To address this, we developed mesoporous organosilica nanoparticles (MONs) coated with hyperbranched polyglycerol (HPG), a hydrophilic, non-immunogenic polymer. The dense, branched HPG inhibits protein corona formation, confirmed by protein assays and SDS-PAGE. HPG-MONs showed reduced macrophage uptake and excellent colloidal stability with high drug loading. They exhibited redox-responsive degradation in glutathione-rich conditions, suggesting efficient clearance and low accumulation. Overall, HPG-coated MONs offer a promising platform for improved drug delivery and immune evasion.
