Abstract
Traditional drug delivery systems for osteoarthritis (OA) face challenges such as rapid joint clearance and limited biocompatibility. Inflammation accelerates cartilage and extracellular matrix degradation, impeding regeneration. To address these issues, we fabricated dCOL2-CM-Cur-PNPs as an OA-specific drug delivery system for anti-inflammation and regeneration. This system comprises curcumin-loaded PLGA nanoparticles (Cur-PNPs) coated with iPSC-derived mesenchymal stem cell membranes (iMSC-CMs) and functionalized with phospholipids targeting damaged type II collagen (dCOL2). The iMSC-CM coating enables sustained drug release and facilitates intracellular uptake by OA chondrocytes. It inhibits M1 macrophages and promotes M2 polarization. dCOL2 enables selective targeting to OA cartilage in vitro and in vivo. dCOL2-CM-Cur-PNPs exhibited significant anti-inflammation and cartilage regeneration in a DMM rat model, highlighting its potential as a next-generation drug delivery platform.
