Bioactive materials and amphiphilic polymer conjugates in the treatment of various diseases

Abstract
We evaluated the potential of an amphiphilic polymer (AP) to bioactive materials such as an antibody or PS. APP and antibody conjugates (APAC) had preserved specificity and structural integrity. APAC reduced aggregation and protected the antibody denaturation or fragmentation from heat and protease stress, subsequently improved binding kinetics and therapeutic efficiency. We then applied AP for the development of photosensitizer (PS) platforms for photodynamic therapy (PDT). AP was conjugated with a PS (APS) to enhance photostability and increases a singlet oxygen generation with targeting effect of PS. APS prevented the decrease in photoactivity of the PS caused by aggregation induced quenching, resulting in a increasing photophysical yield. APS showed superior PDT for treatment of glioblastoma (GBM) in in vivo test. In particular, as co-treated with a checkpoint blockade antibody, APS demonstrates the potential of the synergistic effect as a clinical solution in the treatment of GBM.
