Upconversion nanoparticles loaded injectable supramolecular hydrogels for on-demand glycaemic regulation by optogenetic GLP-1 release

Glucagon-like peptide-1 (GLP-1) receptor agonists have transformed type 2 diabetes and obesity treatment. However, clinical use is hindered by frequent injections and systemic side effects. We present upconversion nanoparticles (UCNPs) loaded in injectable supramolecular hyaluronate (HA) hydrogels for on-demand glycaemic regulation via optogenetic GLP-1 release. Upon transdermal near-infrared (NIR) irradiation, UCNPs emit blue light to activate optogenetic cells, triggering GLP-1 secretion without compromising viability. In mice, using a sweat glucose sensor for continuous monitoring, one subcutaneous injection enabled NIR-triggered regulation of blood glucose levels without inflammation or toxicity. This minimally invasive platform couples durable injectable depots with the temporal precision of light-triggered cell therapy for on-demand glycaemic control.
